ABSTRACT

Objective To detect the influence of rapamycin on the expression of 4 kinds of miRNAs and the effect cell autophagy.To study the relationship of miR-144 and Beclin-1 gene.Methods SKOV-3 cells were treated with 50 ng/mL rapamycin 2 hours and 10 nmol/L 3-methyl adenine 12 hours,the expression of miR-17,miR-20a,miR-144 and miR-155 was detected by RT-qPCR in SKOV-3 cell of different groups,the protein expression of Beclin-1 was detected by Western blot.The targeting effect of miR-144 on Beclin-1 gene was verified by the dual-luciferase reporter assay,Western blot and RT-qPCR.Results The expression of miR-17,miR-144 and miR-155 were in creased compared with NC groups in rapamycin group (P<0.05);miR-17,miR-20a and miR-144 were down regulated compared with NC group in 3-MA group(P<0.05);the protein of Beclin-1 was down expression compared with NC group in rapamycin group.miR-144 could suppress Beclin-1 expression by targeting the specific 3'untranslated region sequence of Beclin-1 gene.Conclusions miR-144 can inhibit the autophagy-related gene Beclin-1 expression and regulate the autophagy process in SKOV-3 cells.

ABSTRACT

As an ancient medicinal and editable resource, pomegranate has a long usage history in the Tibetan medicine, Uyghur medicine and Indian Ayurvedic medicine. Pomegranate pericarp, seeds and flowers are widely used in both TCM and other ethnic medicine. Therefore, it is important to use modern analytical and detective methods to identify the chemical components in order to clarify its material basis and improve the quality control standards. This article summarized the main chemical compositions and quality evaluation methods of pomegranate pericarp, seeds and juice. Analysis was given on questions occurred during the study process. This study provided scientific data for pharmacodynamic material basis of different parts of pomegranate. It also provided scientific references for compre-hensive development and utilization of pomegranate.

ABSTRACT

Objective To study the expression of pericyte and the association between pericyte and angiogenesis in the endometriosis. Methods the endometriosis group is consisted of 20 ectopic endometrium, 20 eutopic endometrium dur-ing proliferation phase, 20 eutopic endometrium during secretory phase of patient with endometriosis. The control group (non-endometriosis ) include 20 normal endometrium during proliferation phase and 20 normal endometrium during prolifer-ation phase of women without entometriosis. The histological morphology parameters such as mean density of microvessel (MVD), pericytes number(PN)as well as the average ratio of pericyte to endothelial cells in microvessels were measured by immunohistochemical and morphological assesses. Results The expression of MVD was lower in the normal endometrium and eutopic endometrium compared with it in ectopic endometrium tissue. The difference of the MVD among the different groups was statistical significant (P<0.05). The expression of MVD in secretory phase endometrium was higher than it in proliferative phase endometrium (P<0.05), and it shows no significant difference between eutopic endometrium and normal endometrium. The expression of PN and PN/MVD in ectopic and eutopic endometrium in EMs group were lower than those in control group. However, we also found that the difference of PN and PN/MVD was not statistically significant. Conclusion Pericytes play an important role in angiogenesis of the endometriosis ,and pericyte coverage contribute to maturing of func-tional vasculature of endometriosis.

ABSTRACT

OBJECTIVE To investigate the antibacterial resistance of clinical isolates from inpatients of non-ICU departments in China in 2006-2007.METHODS Clinical isolates were collected by member hospitals of Mohnarin according to unified protocol.Bacterial susceptibility testing was carried out using Kirby-Bauer(KB),MIC or E-test methods according to CLSI 2006.The data were analyzed by WHONET 5.4 software.RESULTS A total of 77253 clinical isolates from inpatients of non-ICU departments in Mohnarin member hospitals were collected from Jun 2006 to May 2007,of which Gram-negative bacteria accounted for 53549(69.3%),and Gram-positive cocci for 23704(30.7%).Meticillin-resistant Staphylococcus aureus(MRSA) and coagulase-negative Staphylococcus(MRCNS) strains accounted for 57.6% and 83.3%,respectively.There was no glycopeptide-resistant Staphylococcus to be found,but small part of coagulase-negative Staphylococcus was teicoplanin intermediate or resistant.50.9% Of Enterococcus faecalis and 69.9% of E.faecium strains were resistant to high concentration gentamicin.1.1% And 2.7% of E.faecalis and E.faecium isolates were resistant to vancomycin,0.8% and 2.7% resistant to teicoplanin.There were 9.9% isolates of Streptococcus pneumoniae were penicillin-nonsusceptible.About 34.5% Escherichia coli and 23.5% Klebsiella pneumoniae isolates were ESBLs-producing strains.Isolates of Enterobacteriaceae were still highly sensitive to imipenem and meropenem,the average resistant rate was less than 2%.There were about 70% E.coli isolates resistant to quinolones,and 20-60% of non-fermentative Gram-negative bacilli were resistant to antibacterials.CONCLUSIONS Bacterial resistance is severe in inpatients in our country,and the resistant rates are increasing.More effective measures should be taken

ABSTRACT

OBJECTIVE To analyze predisposing factors and investigate the measures to fungemia.METHODS The clinical features,therapy and outcome of 17 patients with fungemia which happened from 1995 to 2004 were(retrospectively) reviewed.RESULTS The morbidity of fungemia was 5.52% in all septicemia in our hospital.Candida(tropicalis) and C.albicans were the major fungal species.There were 6 patients found cultured fungi on local(foci).Tracheotomy and mechanical ventilation (47.06%),indwelling urinary catheter(41.18%),indwelling(central) venous catheter and drainage tube(41.18%),persisted use of broad spectrum antibiotic(76.47%),(chemical) therapy(17.65%),admission to ICU(29.41%),and neutropenia(29.41%) might be the predisposing(factors).The mortality of fungemia was 70.59%.CONCLUSIONS The morbidity of non-C.albicans fungemia is(increasing),preemptic therapy should be begun earlier.Higher dose of fluconazole may be recommended in clinical usage.

ABSTRACT

AIM: To evaluate the safety and efficacy of cefetamet pivoxil vs cefixime in the treatment of acute bacterial infections.  METHODS: A multicenter randomized controlled clinical trial was conducted. Ninety-eight patients (M 43, F 55; age 40 a± s 13 a) with acute bacterial infections of cefetamet group were given cefetamet pivoxil 250-500 mg, po, bid, for 7-14 d, and ninty-five patients (M 44, F 51; age 42 a±14 a) of cefixime group were given cefixime 200 mg, po, bid, for 7-14 d. RESULTS: The overall clinical efficacy rates were 95 % and 94 %, the bacterial clearance rates were 96 % and 94 %, the bacterial sensitive rates were 98 % and 96 %, the adverse reaction rates were 7 % and 6 %, respectively. There was no statistical significance between two groups (P>0.05). CONCLUSION: Cefetamet pivoxil and cefixime are effective and safe in the treatment of acute bacterial infections.
